Abstract
اين مطالعه درباره تلخيص و تعيين خصوصيات بيوشيميايي آنزيم آلفا – آميلاز مقاوم به حرارت و pH اسيدي از Bacillus.Sp.Iranian S1 جدا شده از خاك بيابان ( منطقه گندم بريان دركوير لوت، ايران) است. حداكثرفعاليت و توليد آنزيم در فاز نمايي معادل U ml-1 2.93 در دماي 50 درجه سانتي گراد و pH 5 بود . اين آنزيم توسط رسوب ايزوپروپانول، كروماتوگرافي تبادل يوني در DEAE سلولز DE-52 و ژل فيلتراسيون در سفادكس G-100 خالص سازي شد. در روش انجام شده براي خالص سازي آنزيم آلفا-آميلاز مقدار 98.9 برابر با 25? عملكرد و فعاليت ويژه U ml-1 375.9 از پروتيين به دست آمد .وزن مولكولي آلفا-آميلاز خالص توسط SDS-PAGE در حدود 70 كيلو دالتون تخمين زده شد و آنزيم در طيف گسترده اي از دما و pH به ترتيب بين 30 تا C°110 و pH 3 تا 9 پايدار بود. سنجش با محلول نشاسته به عنوان سوبسترا، فعاليت بهينه آنزيم را در دماي 90 درجه سانتي گراد و pH برابر 5 به معرض نمايش گذاشت . اين آلفا- آميلاز خالص شده و مقاوم به حرارت و اسيديته براي بسياري از اهداف صنعتي مناسب است
Abstract
This study describes the purification and biochemical characterization of a novel thermostable and acidic pH stable ? -amylase from Bacillus sp.Iranian S1 which isolated from the desert soil (Gandom-e-Beryan in Lut desert, Iran). Maximum enzyme production was in exponential phase with activity 2.93 U ml-1 at 50°C and pH 5. The enzyme was purified by isopropanol sedimentation, ion-exchange chromatography on DEAE cellulose DE-52 and gel filtration on Sephadex G-100. The purification procedure performed for ?-amylase enzyme achieved about 98.9-fold purification with 25% yield and specific activity 375.9 U mg-1 of protein. The molecular weight of purified ?-amylase was estimated around 70 KDa by SDS-PAGE and it was stable in a wide range of temperature and pH between 30°C- 110°C and 3-9, respectively. Assayed with soluble starch as substrate, the enzyme displayed optimal activity at 90°C and pH 5. The purified ?-amylase was acid- and thermo-stable with novel properties making it suitable for many industrial purposes.
