Abstract :
End(5′ or 3′)-32P -labelled mRNA coding VEGF (vascular endothelial growth factor) was cleaved by RNase H in the presence of a library of oligodeoxyribonucleotides with random sequences of 20-mer. The most susceptible site, determined by gel electrophoresis and autoradiography, agreed with the most effective antisense target site found with cell-free experiments. Based on these results, the method presented here is indicated to be useful to predict an antisense-effective target site on mRNA.
