Abstract
Ion chromatography (IC) with series bulk acoustic wave (SBAW) detection was developed for monitoring of phosphate, chloride and sulfate during the cultivation of Cephalosporium acremonium and production of cephalosporin C. The advantages of the good selectivity of ion chromatography and the highly sensitive response of SBAW detection have been combined to improve the detection limit. The detection limits (signal:noise=3) for sulfate, chloride, and phosphate were 0.09, 0.02, and 0.07 μg ml−1, respectively. Samples were pretreated by dialysis to remove the medium components which damage the column or impair its performance and cause negative peaks. After dialyzing, the strongly diluted sample can be analyzed using the presented method. The optimization of the chromatographic analysis was investigated. A comparison of IC-SBAW with other methods of IC is made.
