Abstract
The Chiralcel OJ and OJ-R columns were tested for their enantiodiscriminating properties towards cisapride.
Chiral separation of racemic mixtures is of highest importance to the pharmaceutical industry, as the isomers of a given racemate may exhibit substantially differing pharmacological effects, not to mention possible altered toxicity behaviour.
The effect of mobile phase composition upon resolution and analysis time was investigated on both phases by means of an experimental design. Furthermore, a search for useful combinations of variables resulting in acceptable enantioselective separation by simultaneously optimising analysis time and separation quality was aimed at. The method obtained for each column was validated in terms of linearity, detection limit and precision. Both systems were applied to the quantitative determination of cisapride in pharmaceutical preparations.
