Abstract :
Spectrophotometric stability indicating procedures are described for determination of glafenine in pure and dosage forms. First derivative of ratio spectra (1DD) and multivariate calibration chemometric methods are proposed for determination of glafenine in presence of its degradate, glafenic acid. The chemometric methods applied are classical least squares (CLS), multiple linear regression (MLR), principal component regression (PCR) and partial least squares (PLS). These approaches are successfully applied to quantify glafenine using the information included in the absorption spectra of appropriate solutions. In the derivative ratio method, Beer’s law is obeyed in the concentration range of 4–18 μg ml−1 of glafenine, with mean percentage recoveries of (100.03±0.87)% and (99.90±0.95)% at wavelengths 270 and 377 nm, respectively. In multivariate methods, calibration sets of standard samples composed of different mixtures of glafenine and its degradate have been designed. Results were statistically analyzed and compared with those obtained by applying the reference method. They proved both accuracy and precision.
