Abstract :
تنوع ژنتيكي به عنوان يكي از شاخص هاي مهم وضعيت اكولوژيك اكوسيستم هاي آبي مي تواند به عنوان ابزاري منحصر به فرد و توانمند براي ارزيابي و مديريت جوامع زيستي مطرح باشد. به منظور بررسي امكان تنوع ژنتيكي بين گونه هاي كفال طلايي Liza aurata) ) موجود در سواحل جنوب شرقي و غربي درياي خزر با استفاده از روش تعيين توالي ژن D-Loop تعداد 23 نمونه باله ماهي كفال طلايي از استانهاي گيلان (انزلي) و گلستان (گميشان) استحصال گرديد سال انجام آزمايش. DNA نمونه ها با استفاده از روش استات آمونيوم استخراج و كميت و كيفيت آنها با استفاده از روش اسپكتروفتومتري و الكتروفورز ارزيابي گرديد. نمونه هاي DNA تاييد شده ، PCR و سپس توالي يابي شدند. توالي ناحيه D-Loop در DNA ميتوكندريايي كفال طلايي پس از ويرايش شامل 900 جفت باز bp)) بود. با استفاده از نرم افزار MEGA version 5.05 درجه خويشاوندي با تست Tajima 1993بين كفال ماهيان طلايي محاسبه گرديد كه نتايج حاكي از اختلاف ژنتيكي بالا در ميان مناطق گيلان و گلستان بود. براي محاسبه فاصله ي ژنتيكي از آناليز Kimura 2-parameter استفاده شد كه فاصله ي ژنتيكي مشاهده شده بين استانهاي گيلان و گلستان (259/0) بود. ميزان بالايي از Fst بين مناطق گيلان و گلستان وجود داشته كه اين امر بيان كننده ي تمايز بين جمعيت هاي موجود مي باشد. طبق نتايج حاصل از اين بررسي در جنوب درياي خزر ، در محدوده ي استانهاي گلستان و گيلان دو جمعيت متفاوت از ماهي كفال طلايي (Liza aurata) وجود دارد.
Abstract :
Genetic diversity as an important marker of the ecological status of aquatic ecosystems is considered a unique and powerful tool to evaluate biological communities. In order to evaluate the genetic diversity among golden mullet species (Liza aurata) in the southeast and southwest coasts of the Caspian Sea by D-Loop gene sequencing, a total of 23 fin specimens of golden mullet were collected from the Gilan (Anzali area) and Golestan (Gomishan area) provinces. Total DNA from the samples was extracted by ammonium acetate method and the quality and quantity of the extracted DNA were assessed by spectrophotometery and electrophoresis. Polymerase Chain Reaction (PCR) was conducted on the target DNA and then DNA sequencing was carried out. D- loop region in mitochondrial DNA (mtDNA) of golden mullet contained 900 base pairs (bp). Phylogenetic relationships among golden mullet were calculated by MEGA software version 5.05 and divergence time was estimated using Tahjimaʹs test. The results obtained from this study revealed that there were high genetic differences among two regions in the Gilan and Golestan provinces. Kimura 2-parameter was used for genetic distance analysis and the genetic distance recorded between Gilan and Golestan Provinces was calculated at 0.259. The high levels of FsT were observed between Gilan and Golestan Provinces which indicates that genetic differences exist among present populations (p?.05). Based on the results obtained from the south Caspian Sea, probably two different populations of Liza aurata are living in the Gilan and Golestan Provinces.
