مطالعه الگوي پروتئيني تاكي زوئيت نئوسپورا كانينوم توسط SDS PAGE و وسترن بلات در گاوهاي سقط كرده

انگل داخل سلولي نئوسپورا كانينوم در چندين كشور شايع بوده و به عنوان عامل مهمي در سقط جنين و مرده‌زايي در گاوان شناخته مي‌شود. براي مشخص نمودن آنتي‌ژن تاكي زوئيت اين انگل116 نمونه سرم از گاوان سقط نموده در سه ماهه سوم آبستني كه داراي آنتي‌بادي‌هاي ضد نئوسپورا بودند بررسي گرديد. جهت فراهم نمودن محتوي پروتئيني نئوسپورا، تاكي زوئيت خالص انگل ليز گرديد و بعد از الكتروفورز، در غشا نيتروسلولزي جهت رنگ‌آميزي ايمني لكه‌گذاري گرديد. حداقل 9 و حداكثر 13 باند پروتئيني با وزن مولكولي 10 تا 90 كيلو دالتون مشخص گرديد. به نظر مي‌رسد كه تقريبا در سرم تمام گاوهاي سقط نموده دو باند پروتئين با وزن مولكولي 45 و 41 كيلو دالتون، برجسته‌ترين واكنش را در آزمون وسترن بلات نشان مي‌دهند. بر اساس اين مشاهدات اين دو باند پروتئيني مهمترين آنتي‌ژن‌هاي مشاهده شده بعد از وسترن بلات در گاوان ماده سقط كرده مي‌باشند.

The intracellular parasite Neospora caninumis prevalent in several countries and is increasingly recognized as an important cause of abortion and stillbirth in cattle. For characterizeing the tachyzoite antigens of Neospora caninumin aborted cows, sera were obtained from 116 cows which were aborted in the third semester of the pregnancy period and had antibodies to Neospora caninumin ELISA. To obtain the protein content of Neospora, purified tachyzoites were lysed, electrophoretically separated and blotted to nitrocelloluse membrane for immunostaining. Minimum 9 and maximum 13 protein bands ranging from 10 to 90 kDa were observed after immunostaining. It seems that, in almost all of the cows, two protein bands with a molecular weight of 45 and 41 kDa, have a prominent reaction in Western blotting. According to our findings, these two protein bands are the most important antigens observed after Western blotting, in seropositive aborted cows.
