Abstract
تب كيو يك بيماري مشترك انسان و دام است كه در سراسر دنيا رخ داده و توسط باكتري داخل سلولي اجباري كوكسيلا بورنتي ايجاد ميشود. هدف از اين مطالعه بررسي وجود عفونت كوكسيلا بورنتي در گوسفندان سقط كرده در شرق تركيه با استفاده از روش واكنش زنجيرهاي پليمراز بود. تعداد 200 جنين از گوسفندان سقط كرده متعلق به 200 گله از مكانهاي مختلف در بخش شرقي تركيه جمعآوري شدند. نمونههاي ارگان جنيني از قبيل كبد، طحال، ريه و معده گرفته و DNA مربوط به 200 نمونه جمعآوري شده تخليص شد. آناليز واكنش زنجيرهاي پليمراز وجود كوكسيلا بورنتي در ارگانهاي آلوده انجام گرفت، و 4 نمونه (2%) مثبت شدند. علاوه بر اين، سوسپانسيونهاي ارگاني جمعآوري شده به تخم مرغهاي جنيندار تلقيح شده و آناليز واكنش زنجيرهاي پليمراز كيسه زرده، DNA كوكسيلا بورنتي در 5 نمونه (5/2%) را نشان داد. اين مطالعه نشان ميدهد كه عفونت با كوكسيلا بورنتي نقش مهمي در سقط جنينهاي گوسفندي در ناحيه آناتولي شرقي ايفا ميكند.
Abstract
Q fever is a zoonotic disease that occurs worldwide and is caused by the obligate intracellular bacterium Coxiella burnetii. The aim of this study was to investigate the presence of C. burnetii infection in aborted sheep in eastern Turkey using PCR. A total of 200 fetuses were collected from aborted sheep belonging to 200 herds in different locations in the eastern part of Turkey. Foetal organ sles such as liver, spleen, lung and stomach were taken and the DNA was purified from two hundred pooled sles. PCR analysis of C. burnetii presence in infected organs was performed, and 4 sles (2%) were found positive. In addition, the pooled organ suspensions were inoculated to embryonated chicken eggs, and PCR analysis of yolk sacs showed C. burnetii DNA in 5 sles (2.5%). This study shows that C. burnetii infection has an important role in sheep abortions in eastern Anatolia region.
