Abstract :
Objective- To evaluate the effects of autologous platelet-rich plasma (PRP) on wound healing in dogs. Design- Experimental in-vivo study. Animals- 5 mix-breed male adult dogs. Procedures- Under general anesthesia, six 2 × 2-cm, full-thickness skin wounds were created on the back of dogs symmetrically. In each dog, three right side wounds were treated topically with 1.5 ml PRP jelly, whereas left side wounds received no treatment. For macroscopic evaluation, at days 0, 3, 5, 7, 10, 13, 17, 20, and 24, digital photographs were taken from wounds. At days 10, 17 and 24 after wounding, skin biopsies were taken from the center and corner of each wound for hydroxyprolin measurement and histopathologic evaluation respectively. Results- No statistically significant differences were found in percentage of wound contraction, epithelialization and healing between test and control group during study (P>0.05). There were no significant differences between median of hydroxyprolin levels, median of inflammatory cells infiltration, presence of dermal granulation tissue, fibroblast proliferation, arrangement of fibroblasts, collagen deposition and collagen bundle formation scores, in the specimens from left and right wounds (P>0.05). Conclusion and Clinical Relevance- Topical application of platelet-rich plasma can not accelerate repair of the small granulating wounds of dogs.

داﻧﺸﻜﺪه داﻣﭙﺰﺷﻜﻲ، داﻧﺸﮕﺎه ﻓﺮدوﺳﻲ ﻣﺸﻬﺪ، ﻣﺸﻬﺪ، اﻳﺮان. ﻫﺪف ارزﻳﺎﺑﻲ ﺗﺎﺛﻴﺮ ﭘﻼﺳﻤﺎي ﻏﻨﻲ از ﭘﻼﻛﺖ ﻓﻌﺎل ﺷﺪه ﺑﺎ ﺗﺮوﻣﺒﻮﭘﻼﺳﺘﻴﻦ در روﻧﺪ اﻟﺘﻴﺎم ﭘﻮﺳﺖ در ﺳﮓ. ﻃﺮح ﻣﻄﺎﻟﻌﻪ ﻣﻄﺎﻟﻌﻪ ﺗﺠﺮﺑﻲ در ﺣﻴﻮان زﻧﺪه. ﺣﻴﻮاﻧﺎت 5 ﻗﻼده ﺳﮓ ﻧﺮ ﻧﮋاد ﻣﺨﻠﻮط. روش ﻛﺎر- ﺗﺤﺖ ﺑﻴﻬﻮﺷﻲ ﻋﻤﻮﻣﻲ ﺷﺶ ﻋﺪد زﺧﻢ ﺗﻤﺎم ﺿﺨﺎﻣﺖ ﺑﻪ اﻧﺪازه ي 20×20 ﻣﻴﻠﻲ ﻣﺘﺮ در ﻧﺎﺣﻴﻪ ي ﭘﺸﺖ ﺑﻪ ﺻﻮرت ﻗﺮﻳﻨﻪ اﻳﺠﺎد ﺷﺪ. در ﻫﺮ ﺣﻴﻮان زﺧﻢ ﻫﺎي ﻃﺮف راﺳﺖ ﺑﻪ ﻋﻨﻮان ﮔﺮوه درﻣﺎن اﻧﺘﺨﺎب ﺷﺪﻧﺪ ﻛﻪ ﺑﻪ وﺳﻴﻠﻪ ي 1/5 ﺳﻲ ﺳﻲ ژل ﭘﻼﺳﻤﺎي ﻏﻨﻲ از ﭘﻼﻛﺖ درﻣـﺎن ﺷﺪﻧﺪ و ﺑﺮ روي زﺧﻢ ﻫﺎي ﻃﺮف ﭼﭗ ﺳﺘﻮن ﻣﻬﺮه ﻫﺎ ﺑﻪ ﻫﻤﻴﻦ ﻣﻴﺰان ﻣﺘﻴﻞ ﺳﻠﻮﻟﺰ ﻗﺮار داده ﺷﺪ )ﮔﺮوه ﻛﻨﺘﺮل(. ﺑﻴﺴﺖ و ﭼﻬﺎر ﺳـﺎﻋﺖ ﭘـﺲ از اﻳﺠﺎد زﺧﻢ ﻫﺎ درﻣﺎن ﺷﺮوع ﺷﺪ و ﺑﻪ ﺻﻮرت ﻳﻚ روز در ﻣﻴﺎن ﺗﺎ ﺳﻪ ﺑﺎر اداﻣﻪ ﭘﻴﺪا ﻛﺮد. در روزﻫﺎي ﺻـﻔﺮ،3 ، 5، 7، 10، 13، 17، 20 و 24 از ﺗﻤﺎﻣﻲ زﺧﻢ ﻫﺎ ﻋﻜﺲ ﻫﺎي دﻳﺠﻴﺘﺎﻟﻲ ﺗﻬﻴﻪ ﺷﺪ. در روزﻫﺎي 10، 17 و 24 ﺑﻌﺪ از اﻳﺠﺎد زﺧﻢ ﻫﺎ از ﻣﺮﻛﺰ و ﮔﻮﺷﻪ ي ﺗﻤﺎﻣﻲ زﺧﻢ ﻫـﺎ ﻧﻤﻮﻧـﻪ ي ﺑﺎﻓﺘﻲ ﺑﻪ ﺗﺮﺗﻴﺐ ﺑﺮاي ﺑﺮرﺳﻲ ﻣﻴﺰان ﻫﻴﺪروﻛﺴﻲ ﭘﺮوﻟﻴﻦ و ﻫﻴﺴﺘﻮﭘﺎﺗﻮﻟﻮژي ﮔﺮﻓﺘﻪ ﺷﺪ. ﻧﺘﺎﻳﺞ- از ﻧﻈﺮ ﻣﺎﻛﺮوﺳﻜﻮﭘﻴﻚ ﻫﻴﭻ اﺧﺘﻼف ﻣﻌﻨﻲ داري ﺑﻴﻦ دو ﮔﺮوه درﻣﺎن و ﻛﻨﺘﺮل از ﻧﻈﺮ درﺻﺪ ﺗﺸﻜﻴﻞ ﺑﺎﻓﺖ اﭘﻲ ﺗﻠﻴـﺎل، درﺻـﺪ ا ﻧﻘﺒـﺎض زﺧﻢ و درﺻﺪ اﻟﺘﻴﺎم دﻳﺪه ﻧﺸﺪ )p0/05<(. ﻫﻤﭽﻨﻴﻦ ﻫﻴﭻ اﺧﺘﻼف ﻣﻌﻨﻲ داري ﺑﻴﻦ دو ﮔﺮوه درﻣﺎن و ﻛﻨﺘﺮل از ﻧﻈﺮ ﻣﻴﺰان ﻫﻴﺪروﻛﺴﻲ ﭘﺮوﻟﻴﻦ،ﺧﻮﻧﺮﻳﺰي، ﺑﺎﻓﺖ ﻧﻜﺮوزه، ﺗﻮده ﻓﻴﺒﺮﻳﻨﻲ، ﺳﻠﻮل ﻫﺎي آﻣﺎﺳﻲ، ﺑﺎﻓﺖ ﺟﻮاﻧﻪ اي، ﻓﻴﺒﺮوﭘﻼزي وﺟﻮد ﻧﺪاﺷﺖ )p0/05<(. در زﺧﻢ ﻫﺎي ﮔﺮوه درﻣـﺎن ﻣﻴﺰان اﭘﻲ ﺗﻠﻴﺰاﺳﻴﻮن در روزﻫﺎي 10 و 17 ﺑﻴﺸﺘﺮ از زﺧﻢ ﻫﺎي ﮔﺮوه ﻛﻨﺘﺮل ﺑﻮد اﻣﺎ از ﻟﺤﺎظ آﻣﺎري ﻣﻌﻨﻲ دار ﻧﺒﻮد. ﻧﺘﻴﺠﻪ ﮔﻴﺮي و ﻛﺎرﺑﺮد ﺑﺎﻟﻴﻨﻲ- درﻣﺎن زﺧﻢ ﻫﺎي ﺗﻤﺎم ﺿﺨﺎﻣﺖ ﺑﺎ اﻧﺪازه ي ﻛﻮﭼﻚ در ﻧﺎﺣﻴﻪ ي ﭘﺸﺘﻲ ﺳﮓ ﻫﺎ ﺑﺎ ﭘﻼﺳﻤﺎي ﻏﻨﻲ از ﭘﻼﻛﺖ در ﺳﻪ ﻧﻮﺑﺖ ﺑﺎﻋﺚ ﺑﻬﺒﻮد در ﺗﺴﺮﻳﻊ روﻧﺪ اﻟﺘﻴﺎم ﻧﻤﻲ ﺷﻮد.
