Abstract :
Seed priming can restore age-induced loss of seed viability or longevity; however, these invigoration responses may vary with priming agents and seed aging duration. The present study investigated the effect of two potassium salts (K-salts) individually and in combination (1:1) on different lots of muskmelon genotype, MS-1 seeds stored for four consecutive years, from 2013 to 2016 under ambient conditions. The combination of Ksalts significantly enhanced the percent germination of seeds stored for two years (from 2015 to 2016). Further, it also improved root morpho-traits of 10-day-old seedlings. The storage duration had significant effect on the seed surface bacterial and fungal populations. A significantly higher cfu mL-1 microbial counts were recorded for 2013 harvested seeds on three different agar-based media compared to 2014 to 2016 stored seeds. Further, the Scanning EM and FT-IR study revealed the surface microbiological status and functional groups variations, respectively. Thus, aging-related seed coat microflora is responsible for deterioration of the seed coat. Osmo-conditioning cannot restore viability of seeds stored under ambient conditions for more than two years.

آﻣﺎده ﺳﺎزي ﺑﺬر ﻣﯽ ﺗﻮاﻧﺪ ﺻﺪﻣﺎت ﺑﻪ زﯾﻮاﯾﯽ ﯾﺎ ﻣﺎﻧﺪﮔﺎري ﺑﺬر را ﮐﻪ ﻧﺎﺷﯽ از ﺳﺎﻟﺨﻮردﮔﯽ ﺑﺎﺷﺪ ﺗﺮﻣﯿﻢ ﮐﻨﺪ، ﺑﺎ اﯾﻦ ﻫﻤﻪ، واﮐﻨﺶ ﻫﺎي ﺗﻘﻮﯾﺘﯽ ﻣﻤﮑﻦ اﺳﺖ ﺑﺴﺘﻪ ﺑﻪ ﻋﺎﻣﻞ آﻣﺎده ﺳﺎزي )priming agents ( و ﻃﻮل دوره ﺳﺎﻟﺨﻮردﮔﯽ ﺑﺬر ﺗﻐﯿﯿﺮ ﮐﻨﺪ. در ﭘﮋوﻫﺶ ﺣﺎﺿﺮ، اﺛﺮ دو ﻧﻤﮏ ﭘﺘﺎﺳﯿﻢ )K-salt( ﺑﻪ ﻃﻮر ﻣﺠﺰا و ﺑﺎ ﻫﻤﺪﯾﮕﺮ ﺑﻪ ﻧﺴﺒﺖ )1:1( روي اﻧﺒﺎﺷﺘﻪ ﻫﺎي ﻣﺨﺘﻠﻒ از ﺑﺬر ژﻧﻮﺗﯿﭗ ﻃﺎﻟﺒﯽ 1-MS ﮐﻪ در 4 ﺳﺎل ﭘﯿﺎﭘﯽ درﺳﺎﻟﻬﺎي 2013 ﺗﺎ 2016 در ﺷﺮاﯾﻂ ﻣﺤﯿﻄﯽ ﻣﻮﺟﻮد )ambient( اﻧﺒﺎر ﺷﺪه ﺑﻮد ﺑﺮرﺳﯽ ﺷﺪ. ﺗﯿﻤﺎر ﺗﺮﮐﯿﺒﯽ K-salt ﺑﻪ ﻃﻮر ﻣﻌﻨﺎداري در ﺻﺪ ﺟﻮاﻧﻪ زﻧﯽ ﺑﺬر ﻫﺎي اﻧﺒﺎرﺷﺪه در دو ﺳﺎل )از 2015 ﺗﺎ 2016 (را ﺑﻬﺒﻮد ﺑﺨﺸﯿﺪ. ﻫﻤﭽﻨﯿﻦ، اﯾﻦ ﺗﯿﻤﺎر ﺻﻔﺎت ﺷﮑﻠﯽ رﯾﺸﻪ ﮔﯿﺎﻫﭽﻪ ده روزه را ﺑﻬﺘﺮ ﮐﺮد. ﻃﻮل دوره اﻧﺒﺎرداري ﺑﺮ ﺟﻤﻌﯿﺖ ﺑﺎﮐﺘﺮي ﻫﺎ و ﻗﺎرچ ﻫﺎي روي ﺳﻄﺢ ﺑﺬر ﺗﺎﺛﯿﺮ ﻣﻌﻨﺎداري داﺷﺖ. ﻧﯿﺰ، ﺷﻤﺎرش ﻣﯿﮑﺮوﺑﯽ ﺑﺮ ﺣﺴﺐ 1-cfu mLﺑﺮاي ﺑﺬر ﻫﺎي ﺑﺮداﺷﺖ ﺷﺪه در2013 ﮐﻪ روي ﺳﻪ ﺑﺴﺘﺮ ﻣﺘﻔﺎوت ﻣﺒﺘﻨﯽ ﺑﺮ آﮔﺎر ﺑﻮدﻧﺪ ﺑﻪ ﻃﻮر ﻣﻌﻨﺎداري ﺑﯿﺸﺘﺮاز ﺑﺬر ﻫﺎي اﻧﺒﺎر ﺷﺪه 2014 ﺗﺎ 2016 ﺑﻮد. اﻓﺰون ﺑﺮ اﯾﻦ، اﺳﮑﻦ EM و آزﻣﻮن FT-IRﺑﻪ ﺗﺮﺗﯿﺐ ﻣﻮﻗﻌﯿﺖ ﻣﯿﮑﺮوﺑﯿﻮﻟﻮژﯾﮑﯽ ﺳﻄﺢ )ﺑﺬر( و ﺗﻐﯿﯿﺮات ﮔﺮوه ﻫﺎي ﻋﺎﻣﻞ )functional groups( را آﺷﮑﺎر ﮐﺮد . ﺑﻪ اﯾﻦ ﻗﺮار، ﻣﯿﮑﺮوﻓﻠﻮراي ﭘﻮﺳﺘﻪ ﺑﺬر ﮐﻪ ﭘﺪﯾﺪآﻣﺪه از ﺳﺎﻟﺨﻮردﮔﯽ اﺳﺖ ﻣﺴﻮل ﺗﺨﺮﯾﺐ ﭘﻮﺳﺘﻪ ﺑﺬر ﻣﯽ ﺑﺎﺷﺪ. ﻧﺘﯿﺠﻪ دﯾﮕﺮ اﯾﻨﮑﻪ، آﻣﺎده ﺳﺎزي-اﺳﻤﺰي ﻧﻤﯽ ﺗﻮاﻧﺪ زﯾﻮاﯾﯽ)viability( ﺑﺬر ﻫﺎي اﻧﺒﺎرﺷﺪه در ﺷﺮاﯾﻂ ﻣﺤﯿﻄﯽ ﻣﻮﺟﻮد را ﮐﻪ ﺑﯿﺸﺘﺮ از دوﺳﺎل اﻧﺒﺎر ﺷﺪه اﻧﺪ ﺗﺮﻣﯿﻢ ﮐﻨﺪ
