Down Regulated Micro RNA-145 as a Marker to Diagnose Breast Cancer in Egypt

Background: Breast cancer is the most frequently diagnosed malignant neoplasm and is a leading cause of cancer death in female worldwide, Breast Carcinoma (BС) in Egypt shows poor prognostic features, such as higher stage than in developed countries, which may be attributed to different lifestyle and genetic factors. Micro RNA are short, single-stranded, non-protein coding RNA, many types of Micro RNAs are up regulated in cancer state others are down regulated. Aim of the Work: Our aim is to evaluate milt-145 gene expression in serum as a novel biomarker to diagnose breast cancer in Egypt. Methodology: 50 female patients diagnosed as breast cancer by mammography and biopsy. Also 25 age and sex matched females served as a control group. Blood sample were taken from patients and controls serum were separated for detection of miR-145 by q RT-PCR.Results: A significant difference between patients and controls as regard the gene expression of mіRNA-145 (p= A01) was detected in serum. Receiver Operating Characteristic (ROC) Curve analysis showed that the best cutoff value for detection of cancer is miRNA-145=0.795 with sensitivity 74% and specificity 100%. Conclusion: Down-regulation of miRNA-145 gene expression in serum can be used as a biomarker to diagnose breast cancer patient.
