  • Abstract
One of the main diseases of the cardiovascular system, endothelial dysfunction which leads to a diminished NO synthesis and thus increases vessel tone as well as the risk of thrombosis. Chronic inhibition of NO formation is associated with increased blood pressure increases platelets activities impaired fibrinolysis and enhanced thrombin and tissue factor generation. The present work aiming to prove possible prophylactic action of nitric oxide donor against hemostatic changes accompanying nitric oxide deficiency due to chronic hypertension. Material and Methods: Rats were randomly divided into 4 groups equal in number, each contain 20 rats.. Group (1) control rats were given distillate water only. Group (2) hypertension induced group receiving N^G-nitro-L-arginin emethyl ester (L-NAME) added to the drinking water. Group (3) receiving (L-NAME) with drinking water and Nicorandil orally using oral feeding tube (12) Group (4) Rats were received Nicorandil by oral feeding tube. Results: group 2 showed significant increases of mean blood pressure, plasma fibrinogen levels, , significant reduction in mean values of percentages of platelets aggregation, significant increase in mean values of Plasminogen Activator Inhibitor-1 Antigen (PAI-1 antigen), significant increase in mean values of thrombin antithrombin complex (TAT)) and significant increase in mean values of soluble glycoprotein V (sGPV) . Using nicorandil in group 3 can produce significant improve in all hematological parameters measured in group 2. Conclusion, the present study demonstrates, chronic hypotension induced by depletion of nitric oxide can lead to increase in plasma fibrinogen levels, finrinolysis activities as indicated by increase in plasminogen Activator Inhibitor-1 Antigen (PAI-1 antigen), and systemic plasma thrombin which is detected by increases of mean plasma levels of thrombin-antithrombin complex (TAT) and soluble glycoprotein V(sGPV). All these hematological changes can be improved through using nicorandil a nitric oxide donor
