Abstract :
Abstract:Direct redox conversion of microperoxidase-11(MP-11) was studied and demonstrated by cyclic voltammetry on a gold electrode modified by a self- assembled monolayer (SAM) of a new sulfur containing compound (6-Amino-5-Nitroso-4-Hydroxy-1,3-Diaza-5- Cyclohexene-2-Thioone) and DNA film. The results showed a pair of well defined, reversible cyclic voltammetric peaks. It thus obtained at an (MP-11)-S-compound-DNA modified Au electrode. Its formal potential, É0 is 0.224 v for 0.1mol/lit phosphate buffer solution (PBS), pH 7 at a scan rate of 500 mv/s. The formal heterogeneous electron transfer rate constantof MP-11 redox conversion on the S-compound and DNA modified electrode was 2.14 s-1. DNA can be considered as a charge carrier to facilitate electron transfer between the enzyme and electrode.
