Abstract :
Toinvestigatethestabilityanddynamiccharacteristicsofmonolayeradsorbedonunsaturatedlipiddioleoylphosphatidylcholine (DOPC) withvaryingconcentrationsofmyelinbasicprotein(MBP), thesystemisstudiedbyapplyingLangmuirtechniqueandmakingatomicforcemicroscope(AFM)observation,whichisbasedonthemassconservationequationanalysismethodreferredtointhethermodynamicstheory.Asindicatedbysurfacepressure-meanmoleculararea (π−A) and surfacepressure-adsorptiontime(π−T) isotherms,thephysicalpropertiesofmonolayerderivedfromtheinteractionofvaryingconcentrationsofMBPwithliquidcrystallineunsaturatedlipid DOPCmoleculeswerequalitativelystudied.AsrevealedbysurfacemorphologyanalysiswithAFM,themicroregionwasexpandedastheconcentration of MBPinthesubphasewasontheincrease,suggestingthathydrophobicinteractionsledtotheMBPinsertion,thuscausingaccumulationoftheMBPonthesurfaceofthemonolayer. Experimentalresultshavedemonstratedthatthepartitioncoefficientoftheinteractionbetween MBP andunsaturatedphospholipid DOP CandthemolecularareaofMBPadsorbedonthemonolayerfilmwascalculatedusingthemassconservationequation.Inaddition,notonlydoesthevaryingconcentrationofMBPinthesubphaseexertssignificanteffectsonthearrangementandconformationof DOPCmonolayer,italsohascertainguidingsignificancetoexploringthestructuralchangestobiofilmsupramolecularaggregatesaswellasthepathogenesisandtreatmentofrelateddiseases.
