Abstract :
تم الحصول على ثلاث وسبعين عزلة من بكتريا Klebsiella من 200 عينة ادرار مأخوذة من اشخاص مصابين بالتهاب المجاري البولية واظهرت عائدية 4 عزلات (
K38,K33,K27,K44
) للنوع Koxytoca أختبرت حساسية العزلات الأربعة لمضادات الحياة المختلفة فكانت جميع العزلات البكتيرية مقاومة المضادات البيتالاكتام التي تضمنت كل من
G , Penicillin
Penicillin , Amoxicillin , Ampicillin , Benzathin Cephalothin, Piperacillin
Ceftazidim, Cefixime , Cefotaxime , Aztreonam Amoxicillin - Clavulanicacid , Tobramycin
و Clindamycin و Cotrimoxazol و Cephoxitin وكانت جميع العزلات حساسة لمضادات الحياة
Nitrofurantoin و Amikacin و Ticarcillin-Clavulanic acid ، وكانت نصف العزلات حساسة لمضاد الحياة Nalidixic acid و النصف الأخر اظهرت حساسية متوسطة له ، تم تحديد التركيز المثبط الأدنى من مضادات الحياة التي شملت Amoxicillin وPenicilin G و Ampicillin و Piperacillin و
Amoxicillin-Clavulanic acid فكان ولجميع العزلات البكتيرية اكثر من 1024 مايكروغرام /مليليتر بينما كان لمضاد الحياة
Amikacin (32 و16 و4 و2) مايكروغرام أمليليتر للعزلات K38 و K33 و K44 و K27 على التوالي بينما كانت قيمة التركيز المثبط الادنى لمضاد الحياة Nalidixic acid (128و64 و 16 و8) مايكروغرام /مليليتر للعزلات K38 و K33 و K44 و K27 على التوالي
.درست قدرة انتاج البتالاكتاميز من العزلات البكتيرية المنتخبة بطريقة اليود القياسية والطريقة الحامضية القياسية ، واظهرت النتائج أن العزلة
K.oxytoca K38 هي الأغزر انتاجا للانزيم . تضمنت الظروف المثلى لانتاج البيتالاكتاميز من العزلة المنتخبة استخدام وسط نقيع القلب والدماغ وبكمية لقاح بكتيري 5% عند رقم هيدروجيني 7 لمدة 4 ساعات من الحضن بدرجة حرارية
37 مئوية بحاضنة هزازة سرعة اهتزازها 125 دورة في الدقيقة.

Seventy three Klebsiella isolates were obtained from 200 urine sample taken from patients with urinary tract infection. Four of these isolates were identified as K. oxytoca. The sensitivity of bacteria to different antibiotics was tested ,all bacterial isolates were resistant to β-lactam antibiotics included Penicillin G, Benzathin Penicillin, Ampicillin, Amoxicillin, Piperacillin, Cephalothin, Amoxicillin - Clavulanic acid, Aztreonam, Cefotaxime, Cefixime, Ceftazidim, Tobramycin, Clindamycin, Cotrimoxazol, and Cephoxitin. All isolates were sensitive to Nitrofurantoin, Amikacin and Ticarcillin-Clavulanic acid whereas half of the isolates were sensitive to Nalidixic acid and the other half showed an intermediate sensitivity to it . The minimum inhibitory concentration had been determined for Penicillin G, Amoxicillin, Ampicillin, Piperacillin and Amoxicillin -Clavulanic acid it was 1024 microgram/ milliliter for all isolates. Whereas it was (32,16,4,2) microgram /milliliter for the isolates K38, K33, K44 and K27 respectively to Amikacin. It was (128,64,16 and 8) microgram/milliliter for the isolates K38, K33, K44 and K27 respectively to Nalidixic acid .The ability to produce betalactamase from these isolates was studied by the iodometric and acidimetric tests. All isolates produced the enzyme but K.oxytoca isolate K38 was the highly producer. The optimum conditions of betalactamase production from the selected isolate were the use of Brain Heartinfusion broth with 5% of bacterial inoculum at pH 7 for 4 hours of incubation at 37 Co in shaker incubator (125 rpm).
