Abstract :
Cholesterol hyper-accumulation is an increasing risk factor causing cardiovascular diseases. Cardiovascular diseases are leading causes of death worldwide, accounting for 16.7 million deaths annually. An increase from 25.7 to 54.7 million cases of cardiovascular diseases is reported in India. Since 1990, changes in dietary patterns have been the major causes of this condition. However, current treatments such as statins and β-blockers are inadequate and include systemic toxicity such as stent thrombosis and chronic inflammation. Probiotics with cholesterol-lowering ability are ideal safe choices for the prevention of cardiovascular diseases.
Material and Methods: The present study involved culture-dependent method for the enumeration of microorganisms from Pyrrharctia isabella. Microorganisms were isolated and purified and their morphology was studied microscopically. Strains that showed yeast morphology were selected for further probiotic characterization. These isolates were further characterized for probiotic potential based on guidelines from World Health Organization, Indian Council of Medical Research and World Gastroenterology Organisation. The present study assessed cholesterol-lowering potential of the best characterized isolate.
Results and Conclusion: Culture-dependent approach resulted in the isolation of four yeast cultures from the frass of Pyrrharctia isabella caterpillars. Cultures showed antimicrobial activity and other probiotic characteristics such as resisted pH, bile and temperature, similar to the reports by Khisti et al., 2019. However, the novel strain isolated from Pyrrhactia isabella could survive high concentrations of NaCl (1.02M) with an 80.32% survival rate, showing 40.23% hydrophobicity. CP-I culture showed bile salt hydrolase activity as well as susceptibility to sulphatried, penicillin-G, tetracycline and chloramphenicol. This isolate showed the potential to decrease cholesterol levels by 9.16% under in-vitro conditions. Molecular identification of the novel isolate demonstrated that it belonged to Saccharomyces cerevisiae with 97% genetic similarity. Thus, this study demonstrated for the first-time isolation of cholesterol-lowering probiotic yeast Saccharomyces cerevisiae from caterpillar frass under in-vitro conditions.

تجمع زياد كلسترول عامل خطر فزايندهاي است كه موجب بيماري قلبي عروقي ميشود. بيماريهاي 25/ 22 ميليون به 7 / 16 ميليون نفر در جهان مي باشاااد. افزاي 7 / قلبي عروقي عامل مرگ و مير ساااا نه ددود 7 ميليون مورد ابتلا به بيماري قلبي عروقي در هند گزارش شااده اساات. از سااال 1992 ، تغييرات الگوي غذايي عامل اصلي اين وضعيت بوده است. با ايندال، درمانهاي فعلي، مانند استاتينها و مسدود كنندههاي بتا 1 ناكافي ميباشند و ساميت هايي سايساتميم مانند ترمبوز فنر قلب 2 و التهاب مزمن را موجب ميشوند. زيستيارها 3 با توانايي كاه كلسترول انتخابهايي ايدهآل سالمي براي پيشگيري از بيماريهاي قلبي عروقي ميباشند.ﻣﻮاد و روش ﻫﺎ: ﻣﻄﺎﻟﻌﻪ ﺣﺎﺿﺮ ﺷﺎﻣﻞ روﺷﯽ واﺑﺴﺘﻪ ﺑﻪ ﮐﺸﺖ ﺑﺮاي ﺷﻤﺎرش ﻣﯿﮑﺮوارﮔﺎﻧﯿﺴﻢﻫﺎي ﺣﺎﺻﻞ از ﭘﯿﺮﻫﺎرﮐﺘﯿﺎ اﯾﺰاﺑﻼ ﻣﯽﺑﺎﺷﺪ. ﻣﯿﮑﺮوارﮔﺎﻧﯿﺴﻢﻫﺎ ﺟﺪاﺳﺎزي، ﺧﺎﻟﺺﺳﺎزي ﺷﺪﻧﺪ و رﯾﺨﺖﺷﻨﺎﺳﯽ آﻧﻬﺎ ﺑﺎ ﻣﯿﮑﺮوﺳﮑﻮپ ﻣﻮرد ﻣﻄﺎﻟﻌﻪ ﻗﺮار ﮔﺮﻓﺖ. ﺳـﻮﯾﻪ ﻫﺎﯾﯽ ﮐﻪ رﯾﺨﺖﺷـﻨﺎﺳـﯽ ﻣﺨﻤﺮ داﺷـﺘﻨﺪ، اﻧﺘﺨﺎب ﺷـﺪﻧﺪ ﺗﺎ وﯾﮋﮔﯽ ﻫﺎي زﯾﺴﺖﯾﺎري آﻧﻬﺎ ﺗﻌﯿﯿﻦ ﺷﻮد. اﯾﻦ ﺟﺪاﯾﻪﻫﺎ ﺑﺮاﺳـﺎس راﻫﻨﻤﺎي ﺳـﺎزﻣﺎن ﺟﻬﺎﻧﯽ ﺑﻬﺪاﺷﺖ، ﺷﻮراي ﺗﺤﻘﯿﻘﺎت ﭘﺰﺷﮑﯽ ﻫﻨﺪ و ﺳﺎزﻣﺎن ﺟﻬﺎﻧﯽ ﮔﻮارﺷﯽ از ﻧﻈﺮ ﺗﻮاﻧﺎﯾﯽ زﯾﺴـﺖ ﯾﺎري ﺑﺮرﺳﯽ ﺷﺪﻧﺪ. ﻣﻄﺎﻟﻌﻪ ﺣﺎﺿﺮ ﺗﻮاﻧﺎﯾﯽ ﮐﺎﻫﺶدﻫﻨﺪﮔﯽ ﮐﻠﺴﺘﺮول ﺑﻬﺘﺮﯾﻦ ﺟﺪاﯾﻪ را ﻣﻮرد ارزﯾﺎﺑﯽ ﻗﺮار ﯾﺎﻓﺘﻪﻫﺎ و ﻧﺘﯿﺠﻪﮔﯿﺮي: ﻧﺘﯿﺠﻪ روش واﺑﺴـﺘﻪ ﺑﻪ ﮐﺸـﺖ ﺟﺪاﺳـﺎزي ﭼﻬﺎر ﮐﺸﺖ ﻣﺨﻤﺮ از ﭘﻮدر ﮐﺮم اﺑﺮﯾﺸﻢ ﭘﯿﺮﻫﺎرﮐﺘﯿﺎ اﯾﺰاﺑﻼ ﺑﻮد. ﮐﺸـﺖ ﻫﺎ ﻓﻌﺎﻟﯿﺖ ﺿــﺪﻣﯿﮑﺮوﺑﯽ و ﺳـﺎﯾﺮ وﯾﮋﮔﯽ ﻫﺎي زﯾﺴــﺖﯾﺎرﻫﺎ ﻣﺎﻧﻨﺪ ﻣﻘﺎوﻣﺖ در ﺑﺮاﺑﺮ pH، ﺻــﻔﺮا و درﺟﻪ ﺣﺮارت را از ﺧﻮد ﻧﺸـﺎن دادﻧﺪ، ﻣﺸﺎﺑﻪ ﮔﺰارش Khisti و ﻫﻤﮑﺎران در ﺳﺎل 2019. ﺑﺎ اﯾﻦﺣﺎل، ﺳﻮﯾﻪ ﺟﺪﯾﺪ ﺟﺪاﺷﺪه از ﭘﯿﺮﻫﺎرﮐﺘﯿﺎ اﯾﺰاﺑﻼ ﻗﺎدر ﺑﻪ زﻧﺪهﻣﺎﻧﺪن در ﻏﻠﻈﺖﻫﺎي ﺑﺎﻻي ﺳــﺪﯾﻢ ﮐﻠﺮﯾﺪ )1/02M( ﺑﺎ درﺻــﺪ زﻧﺪهﻣﺎﻧﯽ %80/32 ﺑﻮد و %40/23 آﺑﮕﺮﯾﺰي ﻧﺸـﺎن داد. ﮐﺸﺖ CP-I ﻓﻌﺎﻟﯿﺖ آﺑﮑﺎﻓﺘﯽ ﻧﻤﮏ ﺻﻔﺮاوي و ﺣﺴﺎﺳﯿﺖ ﺑﻪ ﺳﻮﻟﻔﺎﺗﺮﯾﺪ، ﭘﻨﯽﺳﯿﻠﯿﻦ-G ، ﺗﺘﺮاﺳﯿﮑﻠﯿﻦ و ﮐﻠﺮاﻣﻔﻨﯿﮑﻞ را ﻧﺸﺎن داد. در ﺷﺮاﯾﻂ ﺑﺮون ﺗﻨﯽ4، اﯾﻦ ﺟﺪاﯾﻪ ﺗﻮاﻧﺎﯾﯽ ﮐﺎﻫﺶ ﺳﻄﺢ ﮐﻠﺴﺘﺮول ﺗﺎ % 9/16 را داﺷـﺖ. ﺷـﻨﺎﺳـﺎﯾﯽ ﻣﻮﻟﮑﻮﻟﯽ ﺟﺪاﯾﻪ ﺟﺪﯾﺪ ﺗﻌﻠﻖ آن ﺑﻪ ﺳـﺎﮐﺎروﻣﯿﺴﺲ ﺳﺮوﯾﺰﯾﻪ ، ﺑﺎ %97 ﺗﺸﺎﺑﻪ ژﻧﺘﯿﮑﯽ، را ﻧﺸﺎن داد. از اﯾﻦرو، اﯾﻦ ﻣﻄﺎﻟﻌﻪ ﺑﺮاي اوﻟﯿﻦ ﺑﺎر ﺟﺪاﺳـﺎزي ﻣﺨﻤﺮ زﯾﺴـﺖ ﯾﺎر ﮐﺎﻫﺶدﻫﻨﺪه ﮐﻠﺴﺘﺮول ﺳﺎﮐﺎروﻣﯿﺴﺲ ﺳﺮوﯾﺰﯾﻪ از ﭘﻮدر ﮐﺮم اﺑﺮﯾﺸﻢ در ﺷﺮاﯾﻂ ﺑﺮونﺗﻨﯽ را ﻧﺸﺎن داد.
