  • Abstract
    In this study, in vitro germination and regeneration ability of endemic Astragalus schizopterus Boiss. was investigated. As we encounter germination handicap, the seeds of A. schizopterus plant,were subjected to sandpaper, boot with a scalpel, or hydrochloric acid (HCl) treatments. The highest germination rate (96.7%) was obtained from seeds plotted with a scalpel. Leaf and petiole explants taken from seedlings grown in aseptic conditions, were cultured in media supplemented with 2.0, 4.0, 8.0 mg/l 2,4-D x 0.5 mg/l KIN containing MS or B5 media, 0.0, 0.5, 1.0, 2.0, 5.0 mg/l 2,4-D x KIN containing MS media, 2.0, 4.0 mg/l IAAx0.5 mg/l and 0.5 mg/l ZEA, l.0 IAAx2.0, 4.0 mg/l ZEA containing MS nutrient environments, hypocotyl explants were cultured in 1.0, 2.0, 4.0 mg/l 2,4-Dx0.5 mg/l BAP or KIN in MS containing environments, adventitious shoot regeneration or somatic embryos were cultured for stimulation. As a result, mostly soft or hard bodied and non-embryogenic callus tissues were obtained from the explants taken from A. schizopterus. There was no shoot regeneration from the callus in any environment, although root formation was observed in some environments.
