Abstract
Bacterial leaf streak of wheat caused by Xanthomonas translucens can cause important losses on wheat in suitable conditions. In order to determine the occurrence of the disease agent on various samples of seed sown widespread 36 wheat cultivars were investigated. For the identification and isolation of the pathogen growth on NA, YDCA, KB, SX and XTS media and at 35 °C, hydrolysis of aesculin, production of hydrogen sulphide from cystein, action lithmus milk, urease activity, arginine dihydrolase, acid production from oxidase and arabinose tests were used. The wheat cultivar Gerek 79 was used as susceptible cultivar for pathogenicity and typical leaf symptoms were obtained. Detection of the pathogen by PCR, T1 and T2 primers which amplified a specific 139-bp fragment were used. With the examination of seed samples of 72 seed samples out of 265 were found contaminated by the pathogen. Xanthomonas translucens was the most and least frequently isolated form Gerek79 (63.63%) and Yıldız 98 (6.25%) wheat cultivars, respectively. General percent infestation ratio of the agent was determined as 27.16% in all of the wheat seeds.
