Enhanced Detection of Prune Dwarf Virus in Peach Leaves by Immunocapture-Reverse Transcription-Polymerase Chain Reaction with Nested Polymerase Chain Reaction (IC-RT-PCR Nested PCR)

To improve the sensitivity of detection of Prune dwarf virus (PDV) in peach trees, the immunocapture-reverse transcription-polymerase chain reaction combined with nested PCR (IC-RT-PCR nested PCR) was modi®ed. The nested PCR substantially improved PDV detection compared with double antibody sandwich-ELISA. Both assays use the same tissue extracts.
