Purification of alkaline phosphatase from chicken intestine by expanded-bed affinity chromatography on dye-linked cellulose

Alkaline phosphatase from chicken intestine was purified on dye-linked cellulose beads. The various dye-linked cellulose beads were screened in the batch mode for purification of the enzyme and the promising dyes were tried in the expanded-bed mode. The onestep expanded-bed affinity chromatography on Cibacron Blue-linked cellulose beads yielded 48-fold purification with an activity recovery of 70%.
