Abstract :
PURPOE: To compare the effect of latanoprot and timolol-XE on ocular preure and perfuion in healthy adult, with repect to epicleral venou preure.
METHOD: A double-maked, placebo-controlled croover tudy of weeklong bedtime treatment with one drop of drug, with placebo contralaterally, followed by a 3-week wahout and alternate-drug/contralateral-placebo repeat. Intraocular preure wa meaured by applanation and by pneumotonometry, providing pulatile ocular circulatory etimate. Meaurement of epicleral venou preure were obtained (Friberg method).
REULT: Twenty ubject participated (five men, 15 women; mean age, 39 year (range, 21 to 55 year); mean baeline intraocular preure, 13.4 mm Hg). A greater decreae in intraocular preure wa een among thee ubject the morning after initiating treatment with latanoprot (−2.0 mm Hg; P < .0001) than with timolol-XE (−0.9 mm Hg; P = .051) (latanoprot veru timolol ΔP = .008). Thi ocular hypotenive effect remained ignificant that evening with latanoprot (−3.2 mm Hg; P < .0001) but not with timolol XE (−1.0 mm Hg; P = .2). By the morning of day 8, mean intraocular preure remained 3.2 mm Hg below baeline with latanoprot and 2.3 mm Hg below baeline with timolol-XE (P < .0001 for both drug). Neither drug altered epicleral venou preure. Among a ubgroup of nine ubject with comparable intraocular preure reduction with the two drug, latanoprot treatment wa aociated with a 16.7% increae in mean pulatile ocular blood flow (P = .04) through the weeklong treatment interval, conitently higher than during timolol-XE treatment of the ame ubject.
CONCLUION: Latanoprot caued an overnight decreae in intraocular preure in normotenive normal eye, and both drug ignificantly reduced intraocular preure within 1 week. Intraocular preure remained higher than epicleral venou preure throughout treatment with both drug. Latanoprot wa aociated with enhanced pulatile ocular perfuion not een with timolol-XE treatment.
