Abstract
PURPOE: To clarify the nature of infiltrating T cell into the anterior chamber of a patient with Behçet dieae.
METHOD: Aqueou humor wa obtained from a patient with ocular Behçet dieae by paracentei. RNA iolated from the cell in aqueou humor wa revere trancribed into complementary DNA. Complementary DNA encoding the variable (V) diverity (D) joining (J) (junctional) region of T-cell receptor β chain V domain (TCR BV) chain wa amplified by T-cell receptor BV family polymerae chain reaction.
REULT: Polymerae chain reaction outhern blot analyi howed that T-cell receptor BV3, BV5, and BV7 were dominantly expreed on ocular T cell of thi patient. In addition, DNA equencing revealed that monoclonal or oligoclonal T-cell accumulation wa found in T-cell receptor BV3+, BV5+, and BV7+ T cell.
CONCLUION: Thee finding ugget that ome T cell infiltrating into the anterior chamber of a patient with ocular Behçet dieae expand by antigen-driven timulation, indicating the pivotal role of T cell in the pathogenei of ocular Behçet dieae.
