Abstract
PURPOE: To determine the mechanim of intraocular preure lowering for the Ocular Hypotenive Lipid, AGN 192024 (Allergan, Inc, Irvine, California).
METHOD: Twenty-five normal human volunteer between the age of 21 and 48 took part in a randomized, double-maked, placebo-controlled, paired-comparion tudy in which intraocular preure, aqueou humor flow, and tonographic reitance to outflow were tudied. Meaurement of aqueou flow were made during the day and at night while ubject lept. Intraocular preure wa meaured with the Goldmann tonometer, and reitance to outflow wa meaured by electronic recording chiötz tonography.
REULT: Intraocular preure wa decreaed by 20% on day 3 in AGN 192024-treated eye in comparion with placebo-treated eye in normal ubject (P < .001). Aqueou humor flow wa timulated 13% during the day (P = .007) and 14% at night (P = .014) by the drug. Tonographic reitance to outflow wa decreaed 26% by AGN 192024 (P < .001), and apparent reitance to outflow (the ratio of intraocular preure to aqueou flow) wa decreaed 31% (P < .001). Auming that AGN 192024 doe not caue prolonged lowering of epicleral venou preure, the reult how that preure-inenitive outflow i enhanced by 50%, wherea tonographic facility of outflow (reciprocal of reitance) wa enhanced 35%.
CONCLUION: AGN 192024 i an ocular hypotenive agent that work by enhancing both preure-enitive and preure-inenitive aqueou humor outflow without diminihing aqueou humor formation
