Abstract :
PURPOE: To tet the utility of DNA Taq polymerae (Taq) treated with 8-methoxyporalen with ultraviolet irradiation and Taq treated with retriction endonucleae in a neted polymerae chain reaction tet for the diagnoi of bacterial endophthalmiti. METHOD: Propective, comparative tudy. Vitreou biopy fluid from 32 cae of clinically diagnoed bacterial endophthalmiti and 10 noninfective control were cultured for aerobic/anaerobic bacteria and teted by neted polymerae chain reaction uing two et of univeral eubacterial primer in triplicate with untreated Taq, Taq treated with 8-methoxyporalen with ultraviolet irradiation, and Taq treated with au 3A1. REULT: Uing untreated Taq, fale-poitive reult were obtained in neted polymerae chain reaction with all 10 control ample, which were not een with the other two method of neted polymerae chain reaction. However, the entivity of neted polymerae chain reaction uing au 3A1 wa the ame enitivity a the conventional culture (34.4%), wherea the enitivity of the neted polymerae chain reaction uing 8-methoxyporalen wa 46.9% higher than in the conventional culture. CONCLUION: To eliminate the problem of fale poitive in bacterial neted polymerae chain reaction, we recommend the routine utilization of Taq treated with 8- methoxyporalen and ultraviolet irradiation.
