Abstract :
Purpoe
To compare the efficacy of a battery of routine and pecial hitologic tain for the detection of acanthamoeba keratiti.
Deign
Obervational tudy.
Method
Nine patient with culture-proven infectiou keratiti whoe clinical differential diagnoi included acanthamoeba and who had undergone penetrating keratoplaty were identified. Three cae each of culture-proven acanthamoeba, fungal, and herpe implex keratiti were reviewed. erial ection of the keratoplaty pecimen were tained with hematoxylin and eoin, periodic acid–chiff (PA), Gomori methanamine ilver (GM), giema, Gram, calcofluor white, and acridine orange. ection were reviewed in a random order and a maked fahion and claified a poitive or negative for acanthamoeba, fungu, or herpe.
Reult
The correct diagnoi wa made by examination of the hematoxylin and eoin tained lide in all cae. Correct diagnoe in decreaing order of frequency were made for lide tained with PA, GM, acridine orange, calcofluor white, giema, and Gram. There were fale-poitive diagnoe made only with calcofluor white and acridine orange tained lide becaue of taining of extracellular debri and other material.
Concluion
When ection are examined by an experienced oberver, hematoxylin and eoin i ueful compared with calcofluor white, acridine orange, GM, PA, giema, and Gram tain for the detection of acanthamoeba keratiti.
