Abstract :
Purpoe
To determine the effect and afety of verteporfin therapy (Viudyne) in Japanee age-related macular degeneration (AMD) patient.
Deign
Open-label, multicenter, propective, noncontrolled tudy.
Method
The etting wa five univerity hopital in Japan. The tudy population wa patient at leat 50 year old, with bet-corrected viual acuity of 20/40 to 20/200 incluive and fluorecein angiography documenting ubfoveal, claic-containing choroidal neovacularization (CNV) econdary to AMD and a leion of greatet linear dimenion ≤ 5,400 μm. For the intervention, patient received verteporfin intravenouly followed by light adminitration 15 minute after the infuion tart. Additional treatment wa given every 3 month through month 9 if leakage from CNV wa oberved on angiogram. The main outcome meaure wa percentage of leion with progreion of claic CNV beyond the area of the baeline leion.
Reult
ixty-four patient were enrolled at five center, and 61 patient completed the tudy. The me-dian number of treatment given during the trial wa three. At the 12-month examination, 12 patient (19%; 95% confidence interval: 11%–31%) had progreion of claic CNV wherea 32 (50%) and 49 (77%) patient had no leakage from claic or occult CNV, repectively. The median viual acuity core (approximate nellen equivalent) from baeline to month 12 increaed from 50.0 (20/100)–56.5 (20/80+2) letter. Fourteen patient (22%) had viual diturbance, including two patient (3%) with acute viion decreae conidered a eriou advere event and one patient (2%) with infuion-related back pain. No photoenitivity reaction were reported.
Concluion
Verteporfin therapy for ubfoveal CNV in thee Japanee patient appeared to have a imilar or better angiographic and viion effect a that oberved in Caucaian patient, with the ame afety profile, upporting it ue in Japanee patient.
