Abstract :
Purpoe
To identify the major oluble protein from human vitreou, and to etablih a baeline for comparion of vitreou ample from eye with variou dieae.
Deign
Laboratory invetigation.
Method
Normal vitreou wa obtained from eight human donor eye and from eight eye of patient undergoing diabetic vitrectomy. Vitreou pecimen were ubjected to D-and MALDI-TOF-M analyi. ix pecific antibodie were ued to identify protein uing Wetern blot. Four protein were localized within ocular tiue in a normal donor eyebank eye.
Reult
We found eight ditinct band on D-in normal vitreou and two additional band (hemoglobin) in eye with diabetic retinopathy. Protein were identified uing MALDI-TOF-M and confirmed by Wetern blot. We etablihed a quantitative analyi of relative protein concentration of undiluted vitreou. Immunohitochemitry localized elected protein within the poterior egment layer.
Concluion
We preent a normal human vitreou protein profile uing current technologie and provide a baeline for comparion to ocular dieae tate. Tiue ditribution of vitreou protein may help to elucidate more pecific protein function.
