Abstract :
Purpoe
To tet whether the partially fluorinated alkane (PFA) perfluorobutylbutane (O44), perfluorohexylethan (O62), and the oligomer OL62HV, recently propoed a artificial vitreou replacement (AVR), have pro-apoptotic effect in rat retinal culture.
Deign
Laboratory invetigation.
Method
Rat retinal cell culture were eeded onto microporou inert to tudy AVR-cell interaction without impairing cell urvival. Cell were treated for 24 hour with O62, O44, and OL62HV. Apoptoi wa analyzed by tranferae-mediated dUTP-biotin nick end-labeling aay and Hoecht tain.
Reult
O44 and O62 did not affect tructural organization and cell urvival in retinal cell culture; however, OL62HV induced increaed apoptoi compared with control culture.
Concluion
OL62HV, a high-vicoity PFA, induce evere retinal damage in human eye, although it uccefully paed animal experimentation. Our in vitro tudy howed a remarkable pro-apoptotic effect of OL62HV and ugget that in vitro tet can contribute to AVR biocompatibility aement.
