Abstract
Primary intraocular lymphoma (PIOL) i a ubtype of central nervou ytem lymphoma. The only currently available animal model of PIOL i a murine model with intraperitoneal inoculation of Rev-2-T-6 lymphoma cell in newborn yngeneic mice. The author now report a new experimental model for PIOL. Rev-2-T-6 cell were inoculated into the vitreou of adult BALB/c mice. Mice were funducopically examined every other day. The mice were acrificed and the eye were tudied by hitology and immunohitochemitry on week 3, 5, 6, 7, or 8 after inoculation. Cytokine mRNA expereion (Il-2, -4, -6, -10, and IFN-γ and CC chemokine receptor-1 (CCR1) wa aayed in the lymphoma cell by microdiection and RT-PCR. IL-10 and -6 level in the vitreou were meaured by ELIA. Within 2 to 4 week, tumor cell from the vitreou migrate through the retina had gathered between the RPE and retina. Rarely, Rev-2-T-6 cell may invade through the RPE into the choroid and clera. Lymphoma cell location wa confirmed by immunohitochemitry. The intraocular lymphoma cell produce high level of IL-10, IFN-γ, and CCR1 trancript. A high level of IL-10 wa detected in the vitreou inoculated with Rev-2-7-6 lymphoma cell. Thi data ugget that RPE cell contitute a barrier of pread of intraocular lymphoma. Intravitreal inoculation of or Rev-2-T-6 lymphoma cell i a novel model of PIOL in immune competent mice and will aide in undertanding the molecular mechanim of the dieae. —Han E. Groniklau
