  • Abstract
    Purpoe To invetigate the effect of exogenou plamin adminitration on the activity of endogenou matrix metalloproteinae-2 (MMP-2) in rabbit and human vitreou. Deign Experimental animal tudy and interventional cae erie. Method Human plamin wa injected into rabbit eye. The active/pro-MMP-2 ratio in vitreou ample wa calculated uing the gelatin zymography. canning electron microcopy (EM) wa performed to oberve the retinal urface. To evaluate the time coure of MMP-2 activity, vitreou ample were collected after the injection of 0.5 IU of plamin, and the active/pro-MMP-2 ratio wa calculated in the ame manner. Immunohitochemical analyi wa performed to confirm the preence of MT1-MMP in the rabbit eye. Human vitreou ample obtained from vitreou urgerie were alo ued for imilar tudie. Reult The active/pro-MMP-2 ratio in the vitreou after the injection of 0.25 IU or 0.5 IU of plamin were ignificantly higher than that of the control (P < .05). EM demontrated that plamin-treated eye howed a mooth retinal urface that wa doe-dependent. Time coure evaluation of the active/pro-MMP-2 ratio in the vitreou after the adminitration of 0.5 IU of plamin found a ignificant difference between the 5 and 15 minute data point compared with that een for the control. Immunohitochemical tudy revealed the preence of MT1-MMP in the inner retina. In human ample, the active/pro-MMP-2 ratio after the plamin injection wa ignificantly higher than the ratio oberved before injection. Concluion Our reult uggeted that activation of endogenou MMP-2 by exogenou plamin i aociated with the induction of poterior vitreou detachment.
