Abstract :
Purpoe
Determine whether antioxidant upplement alter the plama glutathione and/or cyteine redox potential in age-related macular degeneration (AMD) patient.
Deign
Thi wa an ancillary tudy to the Age-Related Eye Dieae tudy (ARED), where ubet of ARED ubject at two ite were tudied at two time point, an average of 1.7 and 6.7 year after enrollment.
Method
Plama glutathione (GH), glutathione diulfide (GG), cyteine (Cy), and cytine (Cy) were meaured by high-performance liquid chromatography, and redox potential of GH/GG (Eh GH) and Cy/Cy (Eh Cy) were calculated. The mean of the metabolite and redox potential were compared by repeated-meaure analyi of variance for ubject receiving antioxidant and thoe not receiving antioxidant.
Reult
At the firt blood draw, the mean for the antioxidant group (n = 153) and no antioxidant group (n = 159) were not ignificantly different for any of the metabolite or redox potential. At the econd draw, the GH parameter were not ignificantly different between the antioxidant (n = 37) and no antioxidant (n = 45) group; however, mean Cy wa ignificantly higher in the antioxidant group (9.5 v 7.2 μmol/l, P = .008). Alo, mean Eh Cy wa ignificantly more reduced in the antioxidant group (−74 v −67.3 mV, P = .03).
Concluion
The ARED antioxidant upplement reduced oxidation of Eh Cy but had no effect on GH. Becaue Cy i important for cell growth, apoptoi, and immune function, the beneficial effect of antioxidant upplementation on progreion to advanced AMD may be partially explained by it effect on Eh Cy and/or it effect on Cy availability.
