Abstract :
Purpoe
To evaluate the polymerae chain reaction (PCR) and equencing-baed riboomal deoxyribonucleic acid (rDNA) typing technique for rapid and enitive diagnoi of fungal keratiti.
Deign
Laboratory invetigation.
Method
The preent tudy included culture-baed method along with PCR and IT2/5.8 rDNA equencing to evaluate fungal infection in 32 corneal craping from patient with upected mycotic keratiti.
Reult
Good correlation wa found between the culture and PCR rDNA equencing-baed method. We alo identified fungal train that were noncultivable in routine culture medium and fungal train, which have been reported carcely in previou report from eatern Indian tudie.
Concluion
The PCR and rDNA equencing-baed approache may be ueful alternative to culture-baed technique for the rapid and enitive diagnoi of fungal keratiti. We found that clinically ignificant pathogenic fungal train can be detected and differentiated by thi method in a hort period. Thi method therefore i ueful for the early treatment of fungal infection.
