  • Abstract
    Purpoe To determine the effect of latanoprot free acid and protaglandin E1 (PGE1) on outflow facility in cultured human anterior egment. Clinical tudie find protaglandin treatment increae uveocleral outflow, but do not agree whether trabecular outflow increae. Cultured anterior egment eliminate the uveocleral pathway and enable a direct aement of trabecular outflow. Deign Laboratory invetigation. Method One anterior egment of an eye pair wa placed in perfuion organ culture and received a continuou infuion of drug while the fellow anterior egment received vehicle. Hitologic change were aeed. Zymography and Wetern blot were ued to analyze matrix metalloproteae (MMP) activity. cleral hydraulic conductivitie were meaured. Reult Latanoprot ignificantly increaed outflow facility (67% ± 11% v control 6% ± 10%, P < .001). Facility change occurred within one hour of receiving drug, reaching a new baeline by 24 hour. Facility change were reverible, requiring about 48 hour to return to pre-drug value. PGE1 caued le facility change (13% ± 17% v control 1% ± 11%, P = .02). Protaglandin treated anterior egment had region of focal detachment and lo of chlemm canal endothelial cell, with lo of extracellular matrix underlying ome area. MMP were not conitently increaed in Wetern blot, zymography, or immunohitochemitry. cleral hydraulic conductivity increaed, but not enough to account for total facility increae. Concluion Protaglandin increae outflow facility in perfuion organ culture of human anterior egment. MMP activity wa not conitently increaed, and cleral hydraulic conductivity wa not increaed ufficiently to account for total facility increae. The hitologic change ugget a direct trabecular mehwork effect.
