Abstract :
Purpoe
To decribe ue of erial len examination to aign cataract phenotype in the Age-Related Eye Dieae tudy (ARED).
Deign
Cohort tudy.
Method
Len photograph were graded annually uing the ARED ytem for claifying cataract. Nuclear grade (0.9 to 6.1) were aigned uing tandard photograph. Percentage of pupillary involvement wa ued to aign cortical and poterior ubcapular grade. Cutpoint were etablihed for the preence or abence of each type of opacity (abent <4.0 for nuclear, <10% for cortical, and <5% of central 5 mm for poterior ubcapular). An algorithm weighted for grade at the lat three examination wa ued to aign cataract phenotype. eparately, cataract phenotype wa aigned a grade predicted at final viit by linear regreion of erial grade. Reult from the two approache were compared and final peron phenotype were etablihed. Thee peron cataract phenotype aignment were compared with phenotype aignment baed on the lat erial grade alone.
Reult
Four thouand ix hundred and twenty-eight ARED participant aged 55 to 80 year at baeline had median follow-up of 10.6 year. Peron phenotype aignment agreed for the two approache in 4,557 (98.5%) participant after ome algorithmic adjudication. Phenotype were no cataract (n = 1,418), nuclear (n = 1,287), cortical (n = 1,396), poterior ubcapular (n = 541), cataract urgery and no pecific opacity type (n = 335), and quetionable (n = 426). Phenotype aignment baed on erial grade and on lat examination alone were in good agreement.
Concluion
erial len photograph obtained over a 10-year period were ued to provide a robut aignment of cataract phenotype. Well-characterized cataract phenotype are of importance a genetic tudie of the ARED cohort are conidered.
