Abstract
Purpoe
To invetigate the Met72Thr (T/C) polymorphim (r1136287) of pigment epithelium-derived factor (PEDF) gene exon 3 in unrelated Taiwan Chinee patient with late age-related macular degeneration (AMD) and control ubject without AMD.
Deign
Retropective cae-control tudy.
Method
We enrolled 190 unrelated Taiwan Chinee patient with late AMD and 90 age- and gender-matched control ubject. Grading of late AMD wa claified baed on a tandardized et of diagnotic criteria etablihed by the International Age-Related Maculopathy Epidemiologic tudy. Late AMD wa claified a either atrophic (dry, grade 4) or neovacular (wet, grade 5). Atrophic AMD refer to dry late-tage AMD without neovacularization, and wet AMD refer to neovacular AMD. Genomic deoxyribonucleic acid wa prepared from peripheral blood obtained from all AMD patient and control ubject. Polymerae chain reaction analyi wa ued to analyze thi polymorphim.
Reult
Of the 190 participant with late AMD, atrophic AMD wa diagnoed in 104 patient and wet AMD wa diagnoed in 86 patient. The genotype ditribution of the Met72Thr (T/C) variant of PEDF wa TT (homozygou T), TC (heterozygou), and CC (homozygou C). The T allele wa found ignificantly more frequently in wet AMD patient than in control (50% v 31%; P =.0005). The allele frequencie in atrophic AMD (30%) and control (31%) did not differ ignificantly (all P = .87). The homozygou T genotype wa more prevalent in wet AMD than in control (26/86 [30%] v nine/90 [10%]; odd ratio, 3.9; all P = .0015). The homozygou T genotype in atrophic AMD patient (8%) and control (10%) did not differ ignificantly (all P = .75).
Concluion
Our data ugget that the PEDF Met72Thr T allele may be a rik factor for wet AMD in the Taiwan Chinee population. PEDF may play a role in the pathogenei of wet AMD.
