Abstract :
Purpoe
To invetigate vacular endothelial growth factor (VEGF) gene polymorphim in unrelated Taiwan Chinee patient with late age-related macular degeneration (AMD) and control.
Deign
Retropective cae-control tudy.
Method
We enrolled 190 late AMD patient and 180 age-matched and gender-matched control. Late AMD wa claified a either dry (atrophic; grade 4) or wet (neovacular; grade 5) according to the International Age-Related Maculopathy Epidemiologic tudy. Genomic deoxyribonucleic acid wa prepared from peripheral blood obtained from all ubject. Polymerae chain reaction were ued to analyze five candidate ingle-nucleotide polymorphim (NP) in VEGF gene: +405C/G (r2010963), −460 T/C (r833061), +674 C/T (r1413711), +936C/T (r3025039), and −2578C/A (r699947).
Reult
Of the 190 late AMD patient, dry AMD wa diagnoed in 104 and wet AMD in 86. Among the five candidate NP tudied, only the +936 C/T wa ignificantly aociated with wet AMD (T allele: 30% in wet AMD v 14% in control; P = 1.45 × 10−5; odd ratio, 2.61; 95% confidence interval, 1.68 to 4.07). No ingle haplotype wa ignificantly aociated with either late AMD or control. Baed on genotype at both VEGF +936 C/T and the complement factor H (CFH) Y402H (r1061170), the aociation of VEGF +936 C/T to AMD wa ignificant when analyzed conditional on the preence of the CFH C rik allele and vice vera (P < .0001). The VEGF +936 C/T wa in trong linkage diequilibrium with CFH Y402H (D′ = 0.99).
Concluion
Both VEGF +936 C/T and CFH Y402H polymorphim are dependently aociated with wet AMD in the Taiwan Chinee population.
