Abstract :
Purpoe To demontrate efficacy and afety of the implantation of neural retinal progenitor cell layer (heet) with it retinal pigment epithelium (RPE) in retiniti pigmentoa (RP) and dry age-related macular degeneration (AMD) patient with 20/200 or wore viion in the urgery eye. Deign Interventional nonrandomized clinical trial. Method Ten patient (ix RP, four AMD) received retinal implant in one eye and were followed in a phae II trial conducted in a clinical practice etting. Early Treatment Diabetic Retinopathy tudy (EDTR) wa the primary outcome meaure. All implant recipient and nine of 10 tiue donor were deoxyribonucleic acid typed. Reult even patient (three RP, four AMD) howed improved EDTR viual acuity (VA) core. Three of thee patient (one RP, two AMD) howed improvement in both eye to the ame extent. Viion in one RP patient remained the ame, while viion in two RP patient decreaed. One RP patient ha maintained an improvement in viion from 20/800 to 20/200 ETDR for more than five year; at the ix-year examination, it wa till maintained at 20/320 while the nonurgery eye had deteriorated to hand motion viion. Thi patient alo howed a 22.72% increae in light enitivity at five year compared to microperimetry reult at two year; the other patient howed no improved enitivity. Although no match wa found between donor and recipient, no rejection of the implanted tiue wa oberved clinically. Concluion even (70%) of 10 patient howed improved VA. Thi outcome provide clinical evidence of the afety and beneficial effect of retinal implant and corroborate reult in animal model of retinal degeneration.
