Abstract :
The study is attempted to investigate the production of lipase from Trichoderma reesei. Lipolytic activity of T. reesei was preliminarily confirmed by zone of clearance on Trybutrin agar medium, lipase production was carried out using modified Reese and Mandals mineral salt solution supplemented with edible oils as substrate in shake flask at 30°C. Fermentation pattern was studied to maximize extracellular lipase production like initial pH of production media, incubation temperature, different substrate, substrate concentration and agitation. The maximum lipase production of 4.23U/ml was obtained after 96hrs of submerged fermentation in a media containing olive oil as a substrate at a concentration of 5% with initial pH of 5.0 and incubated in orbital shaker at 150rpm at 30°C. The extracellular lipase present in the culture media was partially purified by ammonium sulphate precipitation method, characterized for its pH and temperature optima. The optimum pH and temperature on lipase activity were found to be pH 5.0 and 50°C respectively.
