  • Abstract
    بيماري لوك آمريكايي زنبور عسل يا AFB(American foulbrood) به وسيله باكتري گرم مثبت اسپوردار به نام پني باسيلوس لاروا تحت گونه لاروا ايجاد مي‌شود. روش سنتي تشخيص آزمايشگاهي بر اساس تكنيك‌‌هاي كشت، زمان و كار آزمايشگاهي زيادي را مصرف مي‌نمايد. در اين مطالعه با استفاده از باكتري سويه استاندارد و پرايمرهاي اختصاصي، واكنش زنجيره‌اي پليمراز راه اندازي شد و محصول آن روي ژل آگارز 8/0 درصد الكتروفورز گرديد. پرايمرها از ژن 16S rRNA انتخاب شده بودند و يك قطعه bp 700 را تكثير مي‌دادند. حساسيت واكنش زنجيره‌اي پليمراز براي باكتري، اسپور، عسل و لارو آلوده شده به صورت تجربي، تعيين گرديد. كمترين مقدار باكتري و اسپور كه قادر به شناسايي بوديم به ترتيب 28، 31، 330 و 234 cfu/ml تعيين شد. اين روش براي شناسايي پني باسيلوس لاروا لاروا در نمونه هاي عسل، لاروهاي مشكوك به بيماري و يا روي كلني‌هاي رشد كرده روي محيط كشت بدون نياز به آزمايشات بيوشيميايي مي‌تواند به كار رود.
  • Abstract
    American foulbrood (AFB) disease is caused by the sporeforming bacterium Paenibacillus larvae larvae. Traditional diagnosis is based on culture technique is time and laboratory work consuming. In this study with standard strain, PCR was developed by specific primers and PCR products were electrophoresed on 0.8 % agarose gel. The PCR primers were selected on the basis of the 16S rRNA gene and amplify a 700-bp amplicon. Detection limits were determined for suspensions of bacteria and spores and also honey and larvae experimentally contaminated. The lowest number of bacteria and spores that were able to detect were respectively 28, 33, 330 and 243 cfu /ml. This PCR technique can be used to identification of the presence of Paenibacillus larvae larvae spores in honey samples, brood samples or on the colonies that grow on medium.
