Estimate of Callus induction and somatic embryogenesis in cyclamen

ABSTRACT: An efficient protocol for callus induction and somatic embryogenesis of Cyclamen premium Mill. Was developed. In this study we investigate difference between response of two explants type leaf and petiole and two medium salt concentrations MS and ½ MS on callus percentage and somatic embryo number. Media was non-significant on callus production but leaf explants was better than petiole and interaction between explants and media was significant. Leaf explants and MS medium produced the highest callus percentage in cyclamen. Media salt concentration and explants type were significant on somatic embryo number. In ½ MS medium and also in leaf explants the most somatic embryo was obtained. Also interaction between medium and explants was significant and leaf explants produced more somatic embryo in ½ MS medium but petiole explants with MS medium shown highest somatic embryo number. We found for cyclamen callus production leaf explants in MS medium is the best and for highest somatic embryo number in cyclamen Leaf explants and ½ MS is the best.
