Abstract :
Abstract:Leptin is a 16 KD protein that synthesis by white adipose tissue and it involves in regulation of feed Intake, energy balance, fertility and immune function. In cattle, the leptin gene is located on chromosome four. It consists out of 3 exons and 2 introns of which only 2 exon are translated in to protein. We used the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique to screen for DNA polymorphisms of the leptin gene in 70 buffaloes in Shadegan, Ahwaz, Dashte azadegan, Dezfoul and Shoshtar cites (khuzestan provice). In all sample, we amplified a 422 fragment consisting of part of entron 2. The amplified fragment digested with sau3A? restriction endonuclease and subjected to electrophoretic separation in ethidium bromid-strained 2% agarose gel. After enzymatic digestion of the PCR products by sau3A? enzyme generated a 303, 88 and 32 bp fragment of DNA that indicating BB genotype of leptin in buffalo.
