Abstract :
The production of whole plant of Kelussia odoratissima Mozaff. from zygotic embryo culture is an efficient and rapid strategy. Escaping in vitro contamination and long term seed dormancy, seed embryos can be used for mass production of this species. After seeds were sterilized, zygotic embryo was excised and cultured in MS, MS1/2 and B5 basal media. Embryos which were grown in B5 significantly induced root growing and plantlets shape were more natural. By identifying well grown embryos in B5, it was used to determine the most effective nitrogen sources in medium. Medium supplemented with 50 mg L-1 casein hydrolysate and 100 mg L-1 arginine was found more proficient to produce whole plant. By applying 50 mg L-1 casein hydrolysate, embryos fresh weight significantly reached its crescendo whereas 100 mg L-1 arginine induced root growth more efficiently.
