AUergic asthma is a multifac to rial disease, influ en ced by ge netic and envi ronmental
factors, Recent famil y-b ased studies have revealed evidence for linkage o f human
chromosomes 5q31-33, 12q15-24, 11q13 and 15q23.6 as region s likely to contain genes
relat ed to as thma. Among the ca ndida te genes in these regions arc the genes enco di ng fo r
human int erleukin -4, int erleukin-l3 and interleukin-16.
To evaluate thi s linkage, we examined an Iranian population of patients with asthma . A
total o f 30 patients with allergic asthma and 50 normal subjects were studied, Allergic asthma
was co nfirmed using skin prick test and spiro metry, DNA was extrac ted from blood cells
and IT A (-590C(GREATER TTHAN)T) , 11.-13 (R130Q) and IL-1 6 (-295T(GREATER TTHAN) C) pol yrnorphism s were determ ined
by PCR-RFLP method .
O ut o f 30 patients with allergic as thma, the following genotypes for IL-4, 1L-13 and 1L16
cytokincs were fou nd: IL-4 ge no types consisted of 17 (56.7%) CC , 8 (26.7°;(/) CT and 5
(16.7י1.1) TT; IL-13 geno types co ns isted o f 11 (36.7%) G G, 13 (43 ,3%) GA and 6 (20%) AA;
IL -16 geno types consisted o f 21 (76.7%) TT and 7 (23.3%) CT. No patient showed CC
genotype for IL-l 6. A high er p roportion o f case subjec ts with th e C allele for the IL-4, G
allele for the IL-l 3 and T allele for the 1r,-16 polymorphisms was found compared with th e
T , A and C alleles, res pe ctively.
T hese results sugges t an influ en ce o f genetic var iability at th e promo ter of IL-4 gene
(-590C (GREATER TTHAN)T) and a coding region o f IL-13 gene (R130Q) o n the occurre nce o f allergic as thma
and no relationship between IL-16 promote r polymorphism (-295T (GREATER TTHAN) C) and thi s di sease,
