One of the most applicable analgestic drugs which is synthesized from 4- phenyl-piperdine is tramadol[1]. Tramadol is using for help addicted people to quite heroin and its drivatization by effect of central nervous system [2]. Also, tramadol is used as an adjuvant therapy for potentiation of antidepressant drug. Venlafaxine is non-tricyclic antidepressant drug which is represented neuropharmacologic profile distinct from other than existing antidepressants. Inhibiating of neuronal uptake mechanism norepinephrine, serotonin and etc, acts for treating various dispersions conditions[3]. Structural similarities between these two drugs are significant. Both of them contain a methoxy phenyl, a N,N-dimethylamino, and a hydroxyl cyclohexyl group[4]. Simultaneous determination of tramadole and venlafaxine is important due to both of them prescribed for mentally patients. Simultaneous determination of tramadol and venlafaxine has been done by HPLC-MS/MS [5] but this method has high cost, high consumption of solvent, need to great skill, time consuming, etc.In the proposed method, we have determined both of them drugs with at least experimental instruments and partial least square method (PLS) by Parles software. PLS method is used to separate the drugs that overlap together. Tramadol and venlafaxin have excitation and emission wavelengthes close to each other. PLS method was applied for analysis of mixture drugs and evaluation of data. First, one set from mixture drugs was used for training of software and second set was prepared and applied for testing or predicting. In order to evaluation of PLS method for simultaneous determination of tramadol and venlafaxine, another set as evaluation set was prepared and studied by software, too. Predicted concentrations by software were drawn versus actual concentrations for both drugs in mixture. R2 0.9962, 0.9805 was obtained for tramadol and venlafaxine, respectively and relative standard error (RSE) about of 5% indicated capability of PLS method for simultaneous determination of both drugs. Also, LOD was calculated 0.002, 0.001 µg. mL-1 for tramadol and venlafaxine, respectively. Furthermore, precision and accuracy of this method is comparable to HPLC and good recoveries was acquired by both methods.
