Objectives: Bleomycin (BL) is a chemotherapeutic antibiotic used for treatment of cancers in human and small animals. This research study was conducted to investigate the effect of methimazole and the methanol extracted of celery on biochemical changes factors in bleomycin-induced oxidative stress in rats.Materials and methods: Forty adult male Wistar rats (200±20gr) were randomly divided into 4 groups (n=10). In the 5th, 8th and 15th days, bleomycin was administered at 100 mg kg intraperitoneally. The control group (CG) was received normal saline 10 mg / kg for 28 days. The first test group (T1) was received celery hydroalcholic at dose of 400 mg/kg daily. In the T2 group, the rats received BL at dose of 100 mg/kg and methimazole 80mg/kg.The T3 and T4 groups received methimazole and the extract of celery in the same doses, but in the T4 group administration was done after bleomycin injection (don t use as pre-treatment). Finally, the performance of oxidants markers such as MDA in the lung extract and TCA in serum were measured Results: In all groups, malondialdehyde (MDA) concentration was decreased significantly from the control group excluding T4. Total antioxidant concentration (TAC) increased in all test groups but in T3, combination of methimazole and celery (pre- treatment), was significant.Conclusion: The obtained findings illustrated that the methanol extract of celery and methimazole may be efficient in the prevention and treatment of pulmonary fibrosis due to its antioxidant properties. Accordingly, it can be suggested for the prevention side effects of chemotherapeutic drugs use to preference of antioxidant herbal medicines.
