Breast cancer is one of the largest single causes of death among considerable population of women in the world. Due to wide range of chemotherapy and synthetic anticancer drugs side effects, less toxic medications, supported by novel natural products and food supplements, have been widely under scientific investigations and R D studies both by pharmaceutical industries and clinician collaborators. Novel marine natural products have been proven as promising sources of anticancer and antioxidant agents, potent biologically active peptides. The hydrolyzed protein natural product (HPNP) in this study, was extracted from one of the mollusk species of The Persian Gulf of Iran. The aim of this study was to explore the potential anticancer and antioxidant properties of this product by using cellular models. The protein natural product in this study was extracted from one of the mollusk species skin of The Persian Gulf of Iran. After hydrolyzing it with protease to produce bioactive hydrolyzes and fractionating by ultrafiltration, electrophoretic analysis and free amino acid group content were performed. The MTT assay was applied to determine Cell growth inhibitory effect for MCF-7 and MDA-MB-231 (human breast cancer) cell lines. Furthermore, the antioxidant activity measured by ABTS assay and following the treatment, inhibitory effect on matrix metalloproteinase-2 expression and apoptosis were performed. Extend of apoptosis was further assessed by western blotting method. Studies showed that (HPNP) inhibits the cell growth and enhances apoptosis of cells. Cell growth inhibitory effect analysis showed significant increase at higher concentrations (p value 0.05) in treated cells, as compared to untreated.MMP-2 activity demonstrated significant difference between treated and untreated cells (p value 0.05).(HPNP) inhibited cells migration and promote apoptosis through increase in caspase-3 and parp level. Collectively, our results support the anticancer and antioxidant properties of the hydrolyzed protein natural product extract.
