The effect of erythropoietin on hepatocyte apoptotic index following ischemia-reperfusion in the jejunum of cat

Objective: Damages caused by ischemia-reperfusion (I/R) of small intestine are an emergency case which occurs due to various factors such as hernia and twisting of intestines for cats and dogs. Beneficial effects of erythropoietin (EPO) have been demonstrated in various experimental models of ischemic injury but there is no information about the hepatoprotective role of EPO following intestinal I/R in cat. Materials Methods: In this study, 20 tomcats were divided into 4 groups including: 1) sham which had laparatomy operation; 2) jejunum I/R (60/60 minutes) which received normal saline; 3) jejunum I/R which received 1000 IU/kg EPO and 4) jejunum I/R which received 3000 IU/kg EPO via IV after ischemia and before reperfusion. One hour after reperfusion, Liver removed and fixed in in 10% neutral buffered formalin. The tissue samples were processed by routine and standard paraffin embedding and sectioned at 5 μm thickness. Detection of apoptotic cells was performed by nonradioactive in situ end labeling method using TUNEL immunohistochemical technique. Finally, SPSS (ver. 19) software was performed for data analysis. Statistically significant differences among the groups were determined by one-way ANOVA followed by the Tukey post hoc. A P-value of less than 0.05 was considered significant. Results Conclusion: Immunohistochemical results showed that ischemia-reperfusion in the jejunum could induce apoptosis in hepatocytes and increase its apoptotic index compared to sham group (p 0.001). Data analysis also indicated that erythropoietin could inhibit induction of apoptosis in a dose dependent manner (p 0.05). Results revealed that dose of 3000 IU/kg erythropoietin could prevent induction of apoptosis in hepatocytes following ischemia reperfusion in the jejunum of cat, so that reach it to normal level (p 0.05). It can be concluded that administration of erythropoietin can improve side effect of jejunum ischemia-reperfiusion in liver of cat.
