Objective: Regarding to related nephrotoxic mechanisms of gentamicin such as free radicals production and potentiating this effect by furosemide, the aim of present study was evaluation nephrotoxicity induced by gentamicin and furosemide in animal modeling. Material and method: Four groups of rats received normal saline (control group), furosemide, gentamicin, furosemide+ gentamicin, for 8 days. Rats were anesthetized and blood was collected from heart and they euthanized in 9th day. Creatinine, uric acid, urea and NGAL (Neutrophil Gelatinase Associated Lipocalin) was measured in serum. Also, malon dialdehyde, total antioxidant activity and catalase were measured in kidney. Gentamicin significantly increased creatinine, urea and NGAL and renal catalase activity; decreased total antioxidant activity. Above factors did not change by furosemide administration except uric acid and urea. Result: The level of them was significantly increased in group 4 with compared to control group. Co-administration of gentamicin and furosemide induced acute nephrotoxicity. Thus, interaction of gentamicin with furosemide can induced kidney injury probably by stress oxidative reaction.
