Callus Induction via Thin Cell Layer Culture of Two Native Blackberry Genotypes of Iran

In this research, callogenesis was investigated from stem traverse thin cell layer (TTCL) segments of two native genotypes of Iran namely ‘High thorny’ and ‘Low thorny’.For callus initiation, Murashige and Skoog (MS) medium with different types and concentrations of plant growth regulators (PGRs) was used. Results showed that in ‘High thorny’ genotype, the highest callus rate was achieved in MS medium supplemented with naphthaleneacetic acid (NAA), kinetin (KIN) and benzyladenine (BA) (each 1 mg/l). For improving the callus quality, two culture media, MS and Anderson (AN) supplemented with 0.2 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.5 mg/l NAA were employed accompanied with different types of cytokinins.In general, the best callus quality (color, friability) was observed in ‘Low thorny’ genotype on MS medium. In ‘High thorny’ genotype; 2,4-D and NAA had the same effect on callus quality whilst in ‘Low thorny’ genotype NAA was more effective than 2,4-D.
