Objective: Blastocystis hominis is the most common intestinal parasite found in humans and many other hosts in worldwide. Pathogenicity and clinical manifestation of Blastocystis sp. remains controversial and it has been suggested that it may be associated with certain subtypes of organism. The genotype Blastocystis hominis is highly polymorphic. Therefore, a genetic marker would be a powerful tool for the identification or classification of subtypes and could be used as a means to resolve the transmission route or origin of the parasite. The purpose of this study was to evaluate HRM method to identify subtypes of Blastocystis hominis. Material and Method: In this study, 7 mitochondrial gene regions of the Blastocystis hominis were analyzed by bioinformatics. By predicting Insilco methods, the best area was selected to identify subtypes. Suitable primers were designed and synthesized. Results: These reactions were able to identify different subtypes of the parasite after optimization.With a temperature difference of between 0.2 and 1 degree, the different subtypes were well separated. Conclusion: HRM method on mitochondrial genes region was used for the first time to identify Blastocystis hominis subtypes. It seems appropriate techniques on this gene to identify Blastocystis hominis subtypes
